how could I test/prove that the calcium ion is required for the
lectin/carbohydrate complex formation?

You could do a binding experiment inthe presence and absence of a
chelator like EDTA.  This won't prove it is Ca (it could be Mg, or
even Mn), but it is a step in the right direction.
