I am trying to remember which movie contained a scene where the two
main characters get away from two criminals in a bar by playing
pattycake, and by punching the two guys in the face at the end of the
game, instead of hitting their hands together.

Although this type of scene may have occurred in more than one movie,
the one I'm thinking of is probably from the 80's or early 90's.

A pattycake game that ends with punching of bad guys occurs in almost
every "Road" movie made by Bob Hope and Bing Crosby.

Didn't Gene Wilder and Richard Pryor pull this stunt off in one of
their movies from the 70's and 80's? Was it "Stir Crazy"  or "Silver
Streak"?

Pink is right...again... Bob hope and Bing Crosby. It was also done by
Bud Abbot and Lou Costello.

I found one from 1985. The movie "Spies Like Us" starring Chevy chase
and Dan Ackroyd... which is supposedly a tribute to the Hope/Crosby
road pictures.

"Chase and Dan Ackroyd play two low-level CIA agents who are sent on a
rather bogus mission in Afghanistan to basically be expendable decoys.
The boys are dropped inside a giant crate from a plane and when
opened, they are lounging about eating Doritos! So well do they play
their Hope Crosby roles that there is a tribute of sorts to the old
patty-cake game when they encounter a couple of Russian agents."

Bob Hope actually made a short cameo appearance in the movie.

I think it may have also happened in "Who Framed Roger Rabbit".
