What are the 50 largest Yahoo!Groups by membership?

There is no direct method to obtain the list you want.

One can, with some roundabout methods, get a rough idea.

By using site searches of groups.yahoo.com along with 
different figures, you can begin to see where the results
stop showing group memberships and start talking about
other memberships.

10,000 members produces a fair number of groups which 
have reached that goal or exceeded it:

"10000 members" site:groups.yahoo.com
://www.google.com/search?q=%2210000+members%22+site%3Agroups.yahoo.com

Going much higher starts producing less fruitful results.

One of the sites resulting from the search above cites
10,000 members as a thing of the past, in an archived
post in which they note they had 28,900 members in 
December 2002:
http://groups.yahoo.com/group/DailySpell/message/446

But a search of Yahoo Groups reveals their current membership
to be a mere 3724 members:
http://groups.yahoo.com/search?query=dailyspell&submit=Search

On the other hand, a search for 7,000 on Yahoo Groups 
turns up an archived post on the JP1 site which noted
that, at the time, they had surpassed 7,000 members.

A search for JP1 on Yahoo Groups reveals they now have
29,786 members!:

If you keep going like that, you might complete an
authoritative list within a few months.

Hello sublime1,

I agree with sublime1 that there is no way to find this list on
Yahoo!, or with the searches noted by sublime1.  However, you can get
a list of 50, currently, with this search:

Results for:
"activity within 7 days" "members 58000..1000000000" "members * category"
Google
://www.google.com/search?q=site%3Agroups.yahoo.com+%22activity+within+7+days%22+%22members+58000..1000000000%22+%22members+*+category%22

The phrase "activity within 7 days" appears on every Yahoo! Groups
home page.  The phrase "members --" also shows up on every page; I
have filled in the blank with a number range search (indicated by the
two dots) from 58,000 to 1,000,000,000 (because surely no group has
more than 1 billion members, making it a safe number for the high end
of the range).  The low number may have to be revised up or down in
the future, in order to yield exactly 50 results.  I've included the
phrase "members * category" to filter out stray results that include
"members --"  not followed by the word "category."  (The search
produces a few non-matching results at the end.  Perhaps some of the
matching results link to those groups.)

After omitting non-matching results and duplicates, the search produces this list:
