Hi nanny1!!
Drug assays may yield false-positive results caused by cross-reacting
compounds. Compounds that are chemically similar to the test target
compound may cause a positive test result (false positive). This
phenomenon is known as cross-reactivity. The testing products
currently used for urine tests are checked for cross-reactivity by the
manufacturers. The presence of cross-reacting compounds will result in
an increase in the calculated concentration of the sample being
analyzed and therefore influence the incidence of false positive
results.
According to my research Benadryl (diphenhydramine) is not considered
a cross reacting compound for a methadone urine test, not for the most
commonly used these days. It is always included in the NON Cross
Reacting compound list; see for example the following pages from drug
tests manufacturers:
"Urine 1-Drug Cassette - MTD (Methadone) -- Drug Screen Test Cassette
Non Cross-Reacting Compounds":
'Benadryl -- Diphenhydramine -- Non-reactive'.
To see the list open the below page and click on the Compounds link at
the Product Hyperlinks section on the upper right part of the page.

"One Step Multi-Drug Screen Test Card with Integrated E-Z Split Key? Cup":
See at the third page:
"Cross-Reactivity:
A study was conducted to determine the cross-reactivity of the test
with compounds in either drug-free urine or drug positive urine
containing Cocaine, Barbiturates, Benzodiazepine, Amphetamine,
Methamphetamine, Marijuana, Methadone, Methylenedioxymethamphetamine,
Opiate, Phencyclidine or Tricyclic antidepressant. The following
compounds show no cross-reactivity when tested with One Step Drug
Screen Test Card at a concentration of 100 µg/mL."
Again diphenhydramine is listed at the Non Cross Reacting compounds.

"Methadone Drug Test":
Diphenhydramine is in the Non Cross-Reacting Compounds list.
