What steps do you take to identify an unknown bacteria from a sputum
sample of a patient who has a lower respiratory infection?

There are several tests that can be completed to determine the
identityof the unknown microbe.
Gram staining, spore staining, acid fast staining and simple stain ing
can help provide a general idea about the shape of a bacteria and the
general class to which it belongs. Upon completion of thoes test
several tests can be completed to determine the physiological traits
of the unknown culture. For example the mthyl red test, voges
proskauer test, nitrite test, catalase tes, and oxidase test can
detect production of acids, butane diol production, nitrate reduction,
catalase production ad oxidase production. All of these are key in
determining an unknown cultures identity. Also test for lysis of
several bio organic molecules that include casein, urea, fats,
tryptophan. Finally test for utilixation of citrate, and the enzyme
phenlaniline de amminase. Compare your results for the above test with
thoes listed in bergeys manual of determinative bacteriology for the
identification of the unknown culture.
