I have two MS Windows applications that run in "Full Screen" mode only, that I
would like to display on one single screen simultaneously: one above the other.

Is there any way to divide a single screen into "Two Monitors" to do
this?  Does any particular video card support this mode or is there
some other software that could achieve the same result?  Thanks!

Short answer, no. Showing one program above another requires either
two monitors or "tiling" two windows horizontally, which you can 
accomplish by opening two windows, right-clicking the taskbar and
selecting "Tile Windows Horizontally". If you open 2 sessions that
are in fullscreen mode, however, this tiling option is greyed out.
No windows means no tiling. Fullscreen means full screen only.

However, one program will allow you to open two fullscreen sessions.
The only limitation is that you cannot view them simultaneously, as
you asked. The program is freeware, and is called DoubleDesktop:

DoubleDesktop creates a second, full-sized desktop to the right of
your normal one, accessible by clicking an arrow the program puts
in the system tray. On the new desktop, you'll also have a new
taskbar, which will reflect only the programs open on that desktop.
You can open a fullscreen session on the left desktop, and another
on the right, and switch between them using the arrow in the system
tray.

Let me know if this is close enough to suit your needs...

show both running at the same time, one above the other.

If there's a way to tie two monitor outputs into the input for a
single monitor (i.e. with some kind of hardware widget) that would
also be useful ... but it's absolutely got to be on the same screen ;)
